With the added context of India being a transwoman, I understand her comment, since JKR's doesn't believe she is a woman to begin with and even though I disagree with JKR, I know what she's trying to say too (basically citation needed is just like "show me the proof"). Mark Hamill's response is the one that makes absolutely no sense, since there wasn't a discussion or any nuance. I don't understand whatever point he thinks he's making.
